Scutellarin Inhibits the Growth and EMT of Gastric Cancer Cells through Regulating PTEN/PI3K Pathway

Gastric cancer is one of the most common malignancies with a high mortality rate world. This study intends to make clear role and mechanism Scutellarin (Scu), flavonoid isolated from Erigeron breviscapus (Vant.) Hand.-Mazz, in regulating evolvement gastric cancer. We selected different doses Scu treat cells (MGC-803 AGS). Then, cell counting kit-8 (CCK8) assay was conducted verify proliferation tumor cells, while flow cytometry adopted test apoptosis rate. Meanwhile, Western blot examine epithelial–mesenchymal transition (EMT) markers expression phosphatase tensin homolog (PTEN)/phosphatidylinositol 3-kinase (PI3K) apoptosis-related proteins (Bax, Bcl2 Caspase3). Moreover, xenograft experiment nude mice established effect on growth. Furthermore, knockdown model PTEN constructed, influence anti-tumor investigated. As result, inhibited proliferation, EMT promoted dose-dependently. Additionally, attenuated growth vivo. Besides, enhanced reduced phosphorylated level PI3K. mechanistic proved that inactivated PI3K by up-regulating PTEN, thus dampening progression. In conclusion, dampened PTEN/PI3K pathway.
